The one-step seedling formation and rooting mechanism of Acer buergerianum Miq.

Abstract:TheAcer buergerianum,renowned for itsornamental foliageand extensive applications,possesses significantaestheticandeconomic value.To facilitatetherapidproductionofhigh-qualityseedlingsandachieveindustrial-scale propagation,this study employed stem segments with budsas explants to screenforoptimal colection times,sterilization methods,and medium formulations foraxillrybud induction androtinduction.A comprehensive isue culture rapid propagationsystemforAcerbuergerianum was established.Further investigationintotherotformation mechanisminvolvedcytologicalanalysisofadventitiousrootprimordiaandtheexpresionlevelsofkeygenesduringrootdevelopment.Thefindings indicated thata combinationof 75% ethanol disinfection for 2O seconds followed by 0.1% mercuric chloride disinfection for 15 minutes was the most efectivesterilization methodfor explants collected in June.Theoptimal medium foraxillry bud induction was MS+0.5mg/L indole-3-butyric acid(IBA),achieving an induction rate of 82.3% .For adventitious root induction,theoptimal medium was 1/2 MS + 0.6 mg/L IBA,resulting in a rooting rate of 88.7% with an average of 4.9rootsperplant.The initiation and growth of adventitiousroot primordiain Acerbuergerianum werecloselyassociatedwith auxinIBA.At aconcentrationof 0.6mg/L IBA,the formation of root primordia was significantly enhanced,and the relative expression levels of key genes Ab PIN1,AbPIN2,and AbPIN3 during root development increasedmarkedly.Thisstudyprovidesatechnicalreferenceforlarge-scalepropagationofnewvarietiesofAcerbuergerianum and establishes afoundation for exploring its root formation mechanism.
