Genome-wide identification of the RALF gene family in Medicago sativa and expression analysis under salt stress

Abstract:Rapidalkalinizationfactor(RALF)isaclassofsmallpeptides that playcrucial roles inplant growth, development,and stress response.To investigate keygenes involved insalt stressresponse,weidentified membersof the RALF gene family in the whole genome of Medicago sativa and analyzed their response characteristicsunder salt stress.

Throughwhole-genome BLASTanalysis,weidentifiednine RALF genes in M .sativa.The encoded proteins ranged from59to 128 aminoacidsinlength.Their isoelectric pointsclusteredbetween5.13and 5.41.All exhibited hydrophobic properties.These genes were distributed across sixchromosomes,with two pairs exhibiting collinearity. Phylogenetic analysis grouped these genes into four subgroups,and all members retained the conserved motifl and

motif2.Furthermore,promoteranalysisrevealedthatmost MsRALFgenescontainedcis-regulatoryelementsassociatedwith plantgrowth,development,andstressresponse.Undersaltstress,MsRALF1,MsRALF2andMsRALF3expresioly decreased but subsequently increased,peaking at 7d ,whileMsRALF6declined,then increased,and finallydecreased, reaching maximal expressionat3d. MsRALF4,MsRALF5,MsRALF7,MsRALF8 and MsRALF9 all peaked immediatelyon dayO of salt exposure.These findings suggest that the nine MsRALF genes identified in M sativa maybe involved in the plant's response to salt stress,with their expression being regulated by salt stress conditions.
