Analysis of intestinal microbial composition and characteristics of two kinds of snakes in Laotudingzi Nature Reserve

Abstract: This study focuses on the wild Oocatochus rufodorsatus and Gloydius ussuriensis from the Laotudingzi National Nature Reserve. Five adult individuals of each species were sampled,and samples from different sections of the host intestines(small intestine,large intestine,and cloaca) were mixed to explore the differences and characteristics of the overall intestinal microbial composition between the two snake species. High-throughput sequencing of 16S rDNA revealed no significant differences in both Alpha and Beta diversity of the intestinal microbiota between the two species. The dominant bacterial phyla in O.rufodorsatus were Proteobacteria, Firmicutes, Fusobacteria,Bacteroidetes,and Actinobacteria,while those in G.ussuriensis were Proteobacteria, Firmicutes,Fusobacteria,Bacteroidetes,Actinobacteria,and Cyanobacteria.Notably, the abundance of Bacteroidetes showed significant diferences between the two species. At the genus level,significant differences were observed in Salmonella,Citrobacter,Bacteroides,and

Achromobacter. In conclusion,at both the phylum and genus levels,the intestinal microbial community structuresof O rufodorsatus and G .ussuriensis exhibit high consistency. Additionally,numerous probiotics(e. g.,ul14) and potential pathogens(e. g.,Salmonella) were identified in the intestines of both species,providing data support for understanding the composition of wild intestinal microbiota in these snakes.
