Abstract: This study investigated the efect X-ray irradiation on the inhibition lymphocyte proliferation in the blood to determine the optimal irradiation dose the prevention transfusionassociated graft-versus-host disease(TA-GVHD). The experiment was permed at 150kV tube voltage 8mA beam current, the dose was controlled by adjusting the irradiation distance time. The results showed that the inhibition rate lymphocyte proliferation was 92.62%± 1.32% at 40 Gy dose, there was no significant adverse effect on blood routine parameters. When the dose was increased to 70Gy ,although the inhibition rate lymphocyte proliferation was not significantly improved,it led to a sharp increase in the concentration potassium ions in red blood cells,causing hemolysis. This suggests that high doses irradiation can compromise red blood cell integrity. Theree,40 Gy is considered the optimal dose whole blood X-ray irradiation,which effectively inhibits lymphocyte proliferation while maintaining blood component stability. At this dose,it can effectively reduce the risk TA-GVHD without significant impact on blood quality. This study provides important experimental data guidance clinical blood transfusion safety,which is helpful to optimize irradiation parameters,improve the safety effectiveness blood transfusion, reduce the occurrence adverse reactions.
