Abstract:ObjectiveToinvestigatetheoleofapelininegulatingproliferationmigrationandangiogenesisofbladerancer cellsand theposibleregulatorymechanism.MethodsGEOdatabasewasusedtoscreenthediferentiallyexpressedgenes in bladdercancertissues andcells.Bladdercancerandpairedadjacenttissues werecollectedfrom 60 patients foranalysis of apelinexpressionsinrelationtoclinicopathologicalparameters.InculturedbladdercancerJ82celsandumanumbilicalein endothelialcels (HUVECs),theeffectsof transfection withan apelin-overexpressing plasmid orspecificsiRNAs targeting apelin,fibroblastgrowthfactor2(FGF2)andfibroblastgrowthactorreceptor1(FGFR1)onproliferationandmigratioofJ82 cellsandtubetioinUeeadsingateongssayaellsydiogesi changes inFGF2expressionandFGFR1phosphorylationweredetectedusing Westernbloting.ResultsTheexpressionlevelof apelinwassignificantlygherinbadderancertissuestnadjacenttissueandbaddercancercelines(24andJ)also expresed highermRNAand protein levelsof apelinthan SV-HUC-1cels.Apelin expressonlevel inbladdercancer tissues was correlated withtumor inasion,distantmetastasisandadvancedTMstages.Apelinknockdownsignificantlysuessed proliferationandmigrationofJ82celsand decreasedthetotal angiogenic lengthofHUVECs.Incontrast,apelin overexpression significantlypromoted proliferation and migrationand enhancedFGFR1phosphorylation inJ82cels,and increasedthetotalangiogensislengthinUVECsuttisectswereectivelyigatedbytrasfetionoftelswith FGF2 siRNAorFGFR1 siRNA.Conclusion Highexpresionof apelin promotes J82cellproliferationandmigrationand HUVEC angiogenesis by promoting activation of the FGF2/FGFR1 pathway.
