【Abstract】ObjectiveToinvestigatetheefectsofL-theanineonhypoxia-reoxygenation(HR)-inducedinjuryinH9c2 cardiomyocytesanditsuderlyingmechanisms.MethodsAnHRinjuryodelasestablishdinH9c2cardiomyoces.Cellreadomly divided into the following groups:normoxia group,HR group,and HR+L-theanine (2,4,8 mmol/L) groups,and HR + L-theanine(8 mmol/L) + (20 ML385group.Cellviabilityasmeasuredusingcellcountingkit-8assLevelsofreactiveoxygenspeies,alondialdehdeupoide dismutase,cataaecatedrogastiessddacrinsedicali wasdetectedbteialdeoxyucleotidyltrasferas-mdiatedUickndabelinsainngProteinexprsioofcl-asiatedX protein,B-cellp2asps-learactordrelatedfctorndyena-aseedyWstloi MeengerRNAexpresionofuclearfactorerythroid-relatedfactor2andhemeoxygenase-1wasdetectedbyquantitativeeal-tie polymerasechainreactio.ResultsHRstimulationsignficantlyducedcellviabiltyndicreasedtelevelsfocaialijuryarks lactate dehydrogenase,creatine kinase isoenzyme,and cardiac troponin I( P<0. 05 ).Pretreatment with L-theanine effectively restored cell viability and reduced the release of injury markers ( P<0 . 05).HR-induced oxidative stress was characterized by elevated reactive oxygen speciesandmalonddehdelevelsanddecreasedsuperoxidedismutaseandatalaseactivityhichweresignificantlyeversdbyhnine intervention( P<0 .05).Additionally,HR groupshowed significantly increasedapoptosisrateand caspase-3protein expresion,with upregulated-ooraelloaptt ( P<0. 05 ).Furtherstudyfoundthatnuclearfactorerythroid2-relatedfactor2andhemeoxygenase-lexpressonwassignificantly downregulatedinHgoup,whileLeanineintervetionestoredteirexpresiontolevelslosetoomoxiagroup.Thenuclearfactor erythroid 2-related factor2 inhibitor ML385 reversed the above protective effects of L-theanine( P<0.05 ).Conclusion L-theanine attenuates oxidativestresandapoptosisinHc2cardiomycytesxposedtohypoxia-reoxygeationijuryyactivatingteuclearfactoroidrelated factor 2/heme oxygenase-1 pathway.
