Optimization of Extraction Process of Volatile Oil from Fresh Angelica dahurica and Study on Its Components and Antioxidant Activity

Abstract:The volatile oil is extracted from fresh Angelica dahurica by enzyme pretreatment combined with solvent-free microwave extraction (EP-SFME) method. With the yield of volatile oil as the evaluation index, the extraction processof volatile oil is optimized through response surface experiment on the basis of single factor experiment.The chemical components are identified by GC-MS combined with NIST2o.L standard spectral library retrieval,and the scavenging capacity of the volatile oil on DPPH radicals,ABTS radicals, hydroxyl radicals and superoxide anion radicals is studied. The results show that the optimal extraction process is microwave power of 420W , microwave extraction time of 40min ,complex enzyme concentration of 1.5% ,enzymatic hydrolysis time of 2h ,and the ratio of complex enzymes (cellulase:pectinase) of 2:1 . Under these conditions,the yield of volatile oil is (0.27±0.08)% ,which is 47.92% and 17.84% (204号 higher than that obtained by steam distillation(HD) method and solvent-free microwave extraction (SFME) method respectively. A total of 67 components are identified in the volatile oil from Angelica dahurica,mainly including lauryl alcohol, followed by tetradecanol,methyl linolenate,oleyl alcohol, methyl palmitate,9,12-octadecadienoic acid butyl ester, β⋅ -elemene, γ -elemene and so on. The volatile oils obtained by the three methods all exhibit good antioxidant activities,and EP-SFME method is superior to SFME method and HD method.

白芷(Angelicadahurica)为伞形科(Umbelliferae)当归属(Angelica)多年生高大草本植物,是40种大宗药材之一[1],2002年中华人民共和国国家卫生健康委员会将白芷列为药食两用植物,主要用于治疗风寒感冒、眉棱骨痛、牙痛、鼻渊、鼻塞流涕、疮疡肿痛等病症2,也作为一种常见且重要的调味品,在众多菜肴的制作中发挥着关键作用。(剩余13453字)
