Dptimization of High-Density Fermentation Medium and Fermentation Conditionsof Limosilactobacillus fermentum FUA033

Abstract: Limosilactobacillus fermentum FUAo33 converts ellagic acid into urolithin A with multiple biological activities,which is expected to be developed into a new generation of probiotics. This study aims to optimize high-density fermentation medium and fermentation conditions of the strain to reduce the preparation cost of the strain.The medium and fermentation conditions of Limosilactobacillus fermentum FUA033 are optimized by single factor experiment and response surface experiment. The results show that the optimal fermentation medium of Limosilactobacillus fermentum FUA033 is 3.1% glucose, 1.6% peptone, 0.5% (20 K2HPO4 , 0.5% yeast powder, 0.5% beef extract powder, 0.1% Tween 80, (204号 0.5% sodium acetate, 0.2% ammonium citrate, 0.005% manganese sulfate and 0.02% magnesium sulfate. The optimal fermentation conditions are pH of 6.8,temperature of 38°C ,inoculation amount of 2.9% and fermentation time of 20h . The viable bacterial count in the fermentation broth reaches 2.06×1010 CFU/mL,which is 8.77 times that before optimization. The results of this study have laid an experimental foundation for the improvement of viable bacterial count of Limosilactobacillus fermentum FUAo33 direct vat set bacterial agent and the further development and application.
