Genetic diversity analysis of wild tea plant resources based on ISSR markers

Abstract:【Objective】 Shaxian County has abundant resourcesof wildtea plant Camelia sinensis,and conducting phylogeneticanalysisoftearesourcesinthearea hasgreat significance for itsresearch,protection,andutilization. 【Method】Basedon ISSR molecular marker technology,the genetic relationship of 16 wild teaand 5 cultivated resources in Shaxian Countywasanalyzed.NTSYSsoftware wereused for genetic similarityclusteringanalysisand principal coordinate analysis.Result】A totalof9ISSR primers were screened,and88amplifiedbands were detected,including74 polymorphic sites,withapolymorphismrateof84.O9%.Clusteranalysisand principal coordinateanalysis revealed that whenthegeneticcorrelationcoeficientreachesO.68,21tearesourcescanbedivided intothree categories.Rougui'has thefarthest genetic distance fromother species,wild tearesources such as HuyuanandNanyang haveaclose genetic distance from‘Shuixian','Huangdan','Maoxie',and 'Tieguanyin',whileresources inDaluo Townand Dayoushan havea far genetic distance from the reference cultivated species.【Conclusion】 Shaxian County hasabundant wild tea resources, complex genetic diversity,and strong potential for exploring excellent germplasm.
