Establishment and evaluation of an in vitro regeneration system for Sarcopyramisnepalensis

Abstract:【Objective】Thisstudyisintended toexplorea tissuecultureandregenerationsystemfor Sarcopyramis nepalensisand provide technical supportforitslarge-scalesedling production.【Method】Using healthy,disease-free stemsegmentsfromrobustlygrowing plantsasexplants,thesuitabledisinfectionprotocolswerescreened,andthectsof diferentbasicmediaonadventitious bud induction,proliferation,androting were explored.Additionally,theinfluences ofdiferenttransplantingsubstrates,sedlingages,andseedling hardeningdurationsonthetransplantsurvivalrate ofSarcopyramis nepalensis were investigated.【Result】The bestefect isachieved bydisinfecting theexplants with 0.1% (24号 HgCl2 for11 minutes.The most suitable medium foradventitious bud induction is MS +2.0 mg/L 6-BA +O.1 mg/L NAA, withaninduction rate of 84.4% .TheoptimalmediumforproliferationisMS + 0.3mg/L 6-BA + 0.1mg/LNAA,with a multiplicationfactorof 6.12.Therooting culture effctisbestwhenusing1/2MSasthebasic mediumandingO0.5mg/LIBA + 0.1mg/L NAA+1.O g/L activated charcoal,with arooting rate of 100% .The mixture of peat soil and bark ata3:1 ratio proved to be the optimal transplanting substrate,achieving a survival rate of 90.8% and a single-plant growth weight of 0.45g .Transplanting the tissue-cultured seedlings at anage of approximately 6Odays was found to beappropriate,as this stage yielded the highest transplant survival rateand thebest growth performance.Sedling hardening for7days could beterenhance theadaptabilityof the tissue-culturedsedlingsafter transplantationand significantlyimprove the transplantsurvival rate.【Conclusion】This study establishedatisuecultureandregenerationsystem for Sarcopyramis nepalensis.Itprovides technical guidancefor the germplasmresource preservationand industrialized seedling production of Sarcopyramis nepalensis in the future,andalsolaysa foundationforits genetic transformation research.
