Pathogen identification of tomato bacterial stem blight

Abstract:【Objective】Toclarifythe pathogencausing bacterialstem blighton winter tomatoinLianjiang County,Fujian Province,providinga theoretical basis for thecontrolof this disease.【Method】Thepathogen was isolatedandpurified usingthe plate streaking method.Itspathogenicity was confirmed through Koch'spostulates.The pathogen's identity was determined by physiological and biochemical tests,as wellas16S rDNA genesequence analysis.【Result】Theresults showed thatseven bacterial strains were isolatedand purifiedfromdiseased tomato stems,and Koch's postulates confirmed that allseven strains were the pathogens causing this disease.The bacterialcellswere shortrod-shapedand Gram-stain negative.Based on 22 physiological and biochemical tests,such as poly- β -hydroxybutyrate(PHB)production,yellowgren fluorescence productionon King'sB(KB)medium,fructan production,arginine hydrolysis,oxidase test,and utilizationof gucose,xylose,cellobiose,maltose,lactose,trehalose,andglycinebetaineetal,aswellas16SrDNAgene sequenceanalysis,thebacterial strainswere identifiedasPseudomonas viridiflava.【Conclusion】The tomatobacterial stemblightdiseasesinFujianiscausedbyP.viridiflava.
