Analysis of genetic diversity and construction of fingerprinting in Erythrina germplasmusing ISSR molecularmarkers

Abstract:【Objective]This study aimed to evaluate the genetic diversity of Erythrina germplasm,constructDNA fingerprinting,andanalyzephylogeneticrelationshipsusing ISSR molecular markers,therebyproviding amolecular basis forgermplasmresource identification,conservation,and breeding utilizationwithin thisgenus.【Method】Using 26 Erythrinagermplasmsas materials,polymorphic primers werescreened fromISSR primers forPCRamplification.The POPGENE32and NTSYS-pcV2.1O softwares were employed to analyze genetic parameters(Ne,H,I)and perform UPGMAclustering,respectively,andDNA fingerprinting were constructed.【Result】Theresults showedthatatotalof12 ISSR primers with good polymorphism were screened,amplifying168clear bandsacrossthe26 Erythrina germplasms, with a polymorphism rate of 95.18% .Genetic diversity parameters revealed an average effective number of alleles ( Ne )of 1.4423,Nei's gene diversity index( ⋅H )of0.2812,and Shannon'sinformationindex ( I) of0.4423,indicating relatively rich genetic diversity.DNA fingerprinting constructed using primersUBC834,UBC835,and UBC841 coulduniquely identify all26Erythrina germplasms.UPGMAclusteranalysiscategorized the tested materialsintofive majorgroupsata geneticsimilaritycoefficientof O.39.Thisclusteringshowedsomecorrelationwith morphological characteristicsand geographical originbutalsoreflectedpotential geneticexchangeposiblydue tohuman-mediated introduction. 【Conclusion】ISSR molecular markersare efective inrevealing thegenetic diversityofErythrina germplasmandare suitable forresource identificationandphylogeneticanalysis withinthe genus.TheestablishedDNAfingerprinting provide technical support for germplasm identification,variety rights protection,and resource management.
