AbstractObjectiveTo investigate the inhibitory efect of gallic acid (GA)on the antibiotic cross-resistance in Staphylococcus aureus (SA)mediatedbyRhizoma Coptidis (RC)inthis study.MethodsSub-lethal dosesof Paeoniae Radix Rubra extract (PRRE), GA,and the RGA solution, which mixed RC extract (RCE) with GA (1 mL RCE containing 3.6mgGA ),wereused for drug-exposure ofSA for 12 days,respectively.Then susceptibilityanalysis was performed by measuring the minimum inhibitory concentration (MIC)of the induced strains.16S rDNA sequencing was used to identify the induced strains andanalyze their homology.RT-qPCR was used toanalyze the expressionlevelofresistant genes in induced strains to explore the mechanisms of antibiotic cross-resistance.ResultsRPRE,GA,RGAinduced strains could not mediate the antibiotic cross-resistance in SA,respectively. Compared with RCE induced mutantstrins,theexpresinofgnes (smrB,nA,clp,isG,norA,fmtA_lemrB_1,4590,AicaR,BTN44_13995,HMPREF0776_1664 and vraG)in the RGA-induced strains,were down-regulatedby the fold change from 50.00% to 94.16% .Two genes bmr3 and BTN44_15030 were not detected due to their low expression levels. ConclusionsBoth RPR and its active monomer GA could not trigger the cross-resistance to antibiotics in SA,and GAcould inhibit the antibiotic cross-resistance mediated by RCE in SA.
