康替唑胺对宿主天然免疫系统调控作用的相关研究

Abstract Objective To study focus on whether contezolid regulates the host's innate immunity. MethodsThis study was conducted in vivoand invitro.Lipopolysaccharide(LPS)was used to induce endotoxinemia and excessive inflammation in mice,and then the mice were fed with contezolid to observe whether the drug could inhibit the elevationofLPS-induced proinflammatory cytokines and whether it enhanced the expressionof the NRF2/HO-1 pathway.The effect of contezolid on improving the phagocytosis or bactericidal efect of macrophages was also investigated. ResultsThe results showed that low-dose LPS could induce endotoxemia and systemic inflammatory reactions inmice,and contezolid can inhibit the release of proinflammatory cytokines,such as TNF- ∇⋅a and IL-6in the serum of mice,thus inhibiting over-inflammation and having an immune protection effecton the host. The pathological resultsshowed that contezolid could alleviate local congestionand hemorhage in liver and spleen and reduce inflammatorycellinfiltration inducedbyLPS.Moreover,contezolidcould enhance theexpressonof NRF 2/HO-1 pathway.The experiments in vitro implied thatcontezolid reduced the phagocytosis efectof macrophages on Salmonela,but itcansignificantly enhance the bactericidal effect of macrophages.ConclusionContezolidhas an immune regulatory effect on the host's innate immunity,whichcan inhibit theexcessive inflammatoryresponse induced by LPS and enhance the bactericidal effect of macrophages on intracellular bacteria.
