AbstractActinomycin D （Act D） is the first actinomycete antibiotic used for the treatment of malignancies， such as Wilm's tumor of the kidney.The congeners of ActDincludeC-，D-，F-，G-，L-，X-，Y-，and Z-type，which generallyhaveexcellentiologicalactivities，suchasntibacteral，antitumor，ntiiralandant-tberculosisctiiis. Herein，the discovery，biological activities，biosynthesis，and modes of action ofActDagainst tumors，especially NPM1-mutated acute myeloid leukemia，andits congeners have been reviewed in order to facilitate the discoveryof more Act D congeners and synthetic biology studies.
