Effects of Cpx System Gene Deletion on Environmental Response and Immunoprotective Efficacy of Pasteurella multocida

Abstract: CpxA/R,a two-component regulatory system in bacteria, plays an important role in modulating virulence,environmental adaptation,and antibiotic resistance. In this study,the and gene mutants derived from Bovine P . multocida serotype A strain CQ2 were constructed by homologous recombination method. The results showed that the deletion of cpxA and cpxA/R genes had no significant effects on the growth,reproduction,virulence and drug-resistance of strains,and also had no effects on the strains adapting to different metal ion concentrations,oxidative stress,antimicrobial peptide treatment,and nutritional conditions,but it enhanced the strain's tolerance to high temperature,acid stress, and salt stress. Additionally,the deletion of and genes conferred improved cross-protective immunity. In mouse models,the inactivated vaccines derived from the mutants achieved protective efficacy rates of 100% , 50% ,and 50% against bovine P . multocida type A,B and F,respectively,significantly surpassing the protection offered by the wild-type PmCQ2 strain.

Cpx双组份调控系统(以下简称 Cpx 系统)广泛存在于细菌中,由内膜组氨酸激酶CpxA及胞质反应调节剂CpxR 组成[1],当细菌面临复杂环境应激时,位于其内膜上的CpxA 因子发生自磷酸化,将磷酸基团转移到CpxR,促使CpxR蛋白磷酸化,磷酸化的CpxR可作为转录激活因子,从而调控下游相关基因的转录,帮助细菌适应不利环境。(剩余14792字)
