AbstractObjective:Toinvestigate theefectofTianxiangdanonautopaginumanumblicalveinendothelialcels(UVCs)foloing oxidized low-densitylipoprotein(ox-LDL)injury.Methods:HUVECs was divided into control group,ox-LDLgroup,ox-LDL + Tianxiangdan group,and ox-LDL + rapamycin group. activity of HUVECs was detected using the CCK-8 method,the content of nitric oxide(NO) wasmeasuredbycolorimetry,thelevelofautophagywasassessedusingtheMDCmethodautophagicmorphologicalchanges were observedbytransmisionelectronmicroscopeandtheexpresonofautophagrelatedproteinsasdetectedbyWesternBlotethod. Resuts:cellviabitintheox-LDTianxiangdangroupandtheox-LDLrapamycingroupwassignificantlyhighethanthatinthe ox-LDLgroup,and cell permeabityandautophagy levelssignificantly increased in bothgroups( P<0.05 or P<0.01 ).numberof autophagosoeaaiagetofinaingddpsifoatd structuralproteinUNC-51-ikeKinase1(ULK1)andtheautophagproteinmirotubule-asociatedproteinlightchain3(LC3)-/3- increased.Conclusion:Tianxiangdanmayup-regulate ULK1andLC3-I/LC3-Iexpresson levelsbyinhibitingmTORsignal, promote ox-LDL-induced autophagy and improve cell viability.
