探究旋覆代赭汤改善肺癌小鼠顺铂化疗延迟性呕吐的物质基础

【Abstract】Objective: To explore the improvement effct of Xuanfu Daizhe Decoction in the preventive treatment of delayedvomiting after cisplatinchemotherapy in micewith lungcancer.Methods:In vivo biological experiments were conducted to construct a mouse model of lung cancer transplanted tumors.The mice were randomly divided into the model group (cisplatin 0.006g/kg ),the Xuanfu Daizhe Decoction combined with low - dose cisplatin group (0. 006g/kg+1.75g/kg ),the medium- dose group (0.006g/kg+3.5g/kg ),the high-dose group (0.006g/kg+7 (20号 g/kg, ),and he positive group(ondansetron O.O26).Each group was continuously gavaged and cyclicall administered Xuanfu Daizhe Decoction until the eighth day.Cisplatin at a dose of 0.006g/kg was intraperitoneally injected to establisha chemotherapy-induced mouse model of delayed vomiting due to pica.During this period,the body weight and kaolin intakeof the mice were detected.Afterthe experiment,themedullaoblongataand ileum tissues were collected,and the organ indices of the medull oblongata and ileum were calculated.The protein expression levels of 5- hydroxytryptamine 3receptor(5-HT3R)in themedulla oblongata and ileumof the mice were detected by Western Bloting(WB)and immunohistochemistry.Results:In the in vivo experiment,compared with the model group,the Xuanfu Daizhe Decoction significantlyreduced kaolin uptake,significantlydecreased the organ indices of the brain and ileum,and significantly reduced the expression levels of 5-HT3R protein in the medulla oblongata and ileum. Moreover,due totheside efects broughtbycisplatin,thebodyweightofthe miceineach group wassignificantlylower than the initial body weight. Conclusion:Xuanfu Daizhe Decoction may improve delayed vomiting caused by cisplatin chemotherapy by reducing the expression level of 5-HT3R protein.
