【Abstract】Objective：To explore the protective effect of Gei Herba Standard Decoction on rats with ischemic brain injury.Methods：Through the UPLC-Q-TOF-MS technology，the chemical components of Gei Herba Standard Decoction entering the blood and brain were determined.Therat model of ischemic brain injury was constructed by the suture method.SD rats were randomly divided into anormal group，a model group，and groups of high -dose，medium-dose and low-dose Gei Herba（7，3.5，1.75 g/kg），and groups of high -dose，medium-dose and lowdose ellagic acid （150，10，50 mg/kg）．The rats were administered by gavage for7days （onceaday）.The number of cortical Nissl bodieson the ischemic sideofratsand the pathological damage ofrat brain tissue wereobservedrespectively by NISSL and HE staining methods. The levels of TNF - α and 1L-1β in the brain tissue were determined by ELISA. The expressions of proteins such as Bax and Bcl -2 in the brain tissue were detected by Western blot. Results：There was a total of8 components in Gei Herbathat enteredthe blood and brain，among which 5 were brainentering components such as ellagic acid.Compared with that of the model group，the number of Nisl bodies inthe Gei Herba groups and the ellagic acid groups increased，the pathological damage of brain tissue was alleviated，the levels of TNF -α and IL-Iβ in brain tissue decreased， the protein expressions of BDNF，Bcl -2 ，Nrf2 and HIF -1α （20 wereupregulated，and the protein expressions of Bax and GAP43 were downregulated.Conclusion：Gei Herba StandardDecoction hasa protective effctonrats with ischemicbrain injury，and its effect mayberelated to the brainstimulatingcomponentssuchasellagicacid，whichantagonize pro-inflammatory factorsand inhibitcell apoptosis.
