Effects of Solute Carrier Protein-associated Genes on Gastric Cancer Prognosis Prediction and Their Influence on Proliferation, Migration, andInvasion

Abstract: Bioinformaticsanalysis-basedconstructionofaprognosticprediction modeltoinvestigate therelationshipbetween specific solute carier protein (SLC)-related genes and gastriccancer (GC)in immunityand prognosis.RNA sequencing and clinical data were downloaded fromthe cancer genome atlas (TCGA)and gene expressonomnibus (GEO)databases. Diferential expressionofSLC-related geneswas extractedusingRsoftware.UnivariateandmultivariateCoxregression analyses were employed to constructarisk prediction model for GC prognosis basedon SLC-related genes closely associated with survival.The prognostic performance of the model was validated through Kaplan-Meier survivalanalysis,regression analysis,andreceiveroperatingcharacteristic (ROC)curves.Correlationsbetweeriskscoresandimmune-infiltratingcels, tumorimmune microenvironment,andclinical features were assessed.The impactofSLC43A3 knockdownon theproliferation andmigrationofBGC-823cellswas evaluatedvia CCK-8,scratch wound healing,andTranswellassays.Atotalof115 diferentialyexpressed SLC-related genes were identified,with14 genes inked toprognosis.Eight genes wereselectedto establishtheprognosticriskscoring model.TheSLC-relatedgeneprognosticmodeldemonstrateshigh predictivevalueand is significantlyasociatedwithheterogeneityinimmunecellinfiltration,providingimportantreferencesforpersoalized medicationinGCpatients.Functional experiments revealedthatSLC43A3significantlypromotestheproliferation,migration, andinvasionofgastriccancercells.ThisstudymaysuggestnoveltherapeutictargetsforGCtargetedtherapyresearch.
