Cryo-sectioning Technique for Microstructure Studies on Rice Stalks

Abstract: Inorder toobservethemicroscopic structure andlignin distributionofrice stalks efficientlyandconveniently, this studyadoptedthe natural drying-cryo-sectioning methodandsuccessullyobtained thintissuesections.Bycombining the autofluorescencepropertiesofplantsandusing confocalmicroscopy technique,the microscopic structureofrice stalks was observed.This technicalmethodmainlyimprovedthecryosectioning techniqueforricestalks.Aftericestalkswerenaturally dried foracertainperiodof time,they weredirectlyembeddedandsectioned.Sectionscanbeprepared withor withoutsafranin staining toobtainmicroscopicsections withrelativelyintactstructures.ubsequentlyultravioletlight,bluelight,dgen lightwereusedtoexcite theautofluorescenceofricestalks,followedbymicroscopicimagingandphotography.Theresearch results showed that the improved cryosectioning technique could produce tissue sections with a thickness of 1μm and nearly intactcellstructures.Byusingalaserscanningconfocal microscope tocollectredfluorescence signals excitedby greenlight, themicroscopicstructureandlignin distributionofricestalk sectionscanbeobservedquicklyandclearly.Thisstudyprovides references forthe research on the tisse structures and the explorationof the physiological functions inPoaceae plants.
