ABSTRACT:ObjectiveTo investigate the effects of RING 1and YY1binding protein(RYBP),a member of the polycomb group(PcG),on bladder cancer cel proliferation and invasion , as well as the underlying mechanisms involved,so as to provide referencefor the preventionand treatment of bladder cancer.MethodsOverexpresing and knocking down of RYBP were achievedin bladdercancercellines(T24,UM-UC-3,and 5637)via plasmidsor siRNAs.Cell proliferation was assessedvia thiazolylblue(MTT)andcolony formation assys,whereas migrationwas evaluated via sratch and Transwellassys.Changes in epithelial-mesenchymal transition(EMT)markers and otherrelated proteins wereexamined with Western bloting.Results RYBP overexpression significantly inhibited bladercancercell proliferation,invasion,and migration,whereasRYBP knockdown promoted these behaviors.Western bloting resultsrevealed thatRYBP overexpresion downregulatedthe expressionsofEMTmarkers N-cadherin,Vimentin,and Slug,utupregulated the expresionofE-cadherin.Conversely,YBP knockdown upregulated theexpressions of N-cadherin,Vimentin,and Slug,whilereducing theexpressionof E-cadherin. ConclusionRYBPappears toinhibit the proliferation and migrationof bladdercancer cels viatheEMTpathway,indicating its potential application in bladder cancer therapies.
