Construction of pH/ROS Dual-Responsive Microgel Multidrug Co-delivery System

Abstract:Smartresponsivenanocarriers have emergedas apromising strategy for tumor therapy,demonstratingsignificant clinicalpotentialdue totheirhigh targeting capabilityandexcellntbiocompatibility.Anamphiphiliccopolymer, poly(propylene sulfide)-poly(3-acrylamidophenylboronic acid-co-poly(ethylene glycol) methyl ether methacrylate) (PPSk. 1 P(APBAm..co.OEGMAn) ),was designed and synthesized. Through self-assembly,reactive oxygen species (ROS)-responsive polymeric micelles were obtained. The hydrophilic I Λo(APBAm..co.OEGMAn) )shell was further crosslinked using the polyphenolicdrug proanthocyanidin (PC),forming a boronate estercovalent network that enhanced the stabilityof the nanocarier while introducing pHresponsivenessand yielding dual pH/ROS-responsive microgel carriers.The particle size, surface morphology,and crossinking behavior of the microgels were characterized using dynamic light scattring (DLS), scaning electron microscopy (SEM),andultraviolet-visible(UV-Vis)spectroscopy.Thedrug-loadingandreleasebehavior of smal-molecule doxorubicin (DOX)and macromolecular bovine serum albumin (BSA) were investigated,demonstrating eficientdrug encapsulationand stimuli-responsiverelease.Theresultsconfirmedthatthe microgelcarrier exhibited pH/ROS dualresponsiveness,a stable independent loading of multiple drugs,and controled drug release.This study providesa scientific foundation for precision drugdelivery,intellgent release systems,andovercoming multidrug resistance incacer therapy.
