Abstract：ObjectiveToexplorethecorrelationofELFexpresionlevelwithprognosisofcolorectalcanceranditsreulatory roleincolorectalcancercellproliferationandmetastasis.MethodsWeanalyzedtheexpresionlevelsofELFN1across33 cancer typesusingpubliclyavailabledatabasesandidentifieddiferentialgenesrelatedtoELFN1incolorectalcancer.Gene functionannotationandeichmentaalysisereusedtodentifyteinvoledignalingthways.Lgisticaalysisapla MeieranalysisandCoxregressionanalysiswere performed toevaluate thecorelation between ELFN1expressionand clinicopathologicalparametersandsurvivalofcolorectalcancer patients.qPCRand Westernbloting wereused tovalidatethe expresion levelsofELFN1 indifferentcolorectalcancercellinesandtisses，andTranswellandEDU experiments were carriedouttoasesstheeffectof ELFN1 knockdownon biological behaviors of SW480cels.ResultsELFN1 was highly expresed in14cancers，anditsexpresion wassignificantlyhigherincoloncancer issues thaninadjacenttisses.Ahigh expresionofELFN1mRNAwasassociatedwithapooreroverallsurvivalofcolorectalcancerpatients.Coxregresionanalysis indicatedthat ELFN1expression wasan independentprognosticfactorforoverall survival of thepatients.ELFN1 was significantlyenrichedintumor metastasisandproliferationand participated inseveraltumorsignalingpathways.Thecolon cancercellinesshowedsigificantlyigherexpressionlevelsofELFN1thannormalcells，ELFN1knockdownobviously inhibitedproliferationandmigrationofSW480celsinvitro.ConcusionELFN1isoverexpressedincolorectalcancerandis associated withpoorclinicalprognosisof thepatients.AhighELFN1expressionisasociatedwithmalignantphenotypesof colorectalcancerandpromotescancercellproliferationandmetastasis，suggestingitspotentialasaprognosticbiomarkerfor colorectal cancer.
