Moxibustion promotes endometrial repair in rats with thin endometrium by inhibiting the NLRP3/pyroptosis axis via upregulating miR-223-3p

Abstract： Objective Toexplorethemechanism throughwhichmoxibustionpromotes endometrialrepair inrats withinthin endometrium （TE）. Methods Female SD rats were randomized into control group， 95% anhydrous ethanol-induced TE model groupand moxibustion （at "GuanYuan"） group.High-throughputsequencing Was used toidentifythe target genes ofTE，and thetargeting relationshipbetweenmiR-223-3pandNRP3wasverifiedusingadualluciferaseassay.Histopathologicalofrat uterus WasobservedwithHEstainingandexpressonsofmiR-22-3pandNLRP3weredetectedusingRT-qPCR;serumlevels of IL-1βandI-18oftheatsweredetectedusingELIAandproteinexpreionsofNRP3ASCaspase-1andGDMDin theuterusweredetectedwithWesternbloting.Thepregnanciesoftheratsaftertreatmentwerecounted.ResultsEnrichment analysisofthediferentialgenessuggestedup-regulatedinflammatoryresponseinTE，anddualluciferaseassyverified targetedinhibitionofNLRP3expresionbymiR-223-3p.TheratmodelsofTEhadsignificantlydecreasedendometrial thicknessandreducedendometrial glandsandbloodvesselswithenhancedmRNAexpressionofNLRP3，increasedserum levels of IL-1βandIL-18，up-regulated protenexpesionsof3，ASC，caspase-1andGDD，owered pregnancates on both the afectedand unaffcted sides and the overallnumber of pregnancies.Treatment oftherat models with moxibustioobviouslyincreasedtheendometrialthicknessandthedensityof glandsandbloodvessels，up-regulatedmiR-223 ⋅3p expression，loweredserumIL1βandIL-18levelsandtheproteinexpressionsofNP，ASC，caspase-1andGDand significantlyincreased thenumber of pregnancies.Conclusion Moxibustionat "Guan Yuan"acupointup-regulatesthe expresion of miR-23-3p， which results in targeted inhibition of NLRP3 to suppresspyroptosis and promote endometrial repair inratmodelsof TE.
