Electroacupuncture combined with rehabilitation training improves neurological function of mice with cerebral ischemia by promoting astrocyte transdifferentiation

Abstract:Objective To exploretheeffectsofacupuncture combined withrehabilitation training forpromoting transdiferentiationofastrocytes intoneuronsinmiceaftercerebralischemia.Methods MaleC57BL6Jmice weresubjectedto intracerebralmicroinjectionof anadeno-assciated viruscarrying the GFAPpromoterfor NeuroD1and Ngn2 overexpression intheastrocyes,folowed3or1dayslaterbylectrocoagulationofthedistalmiddleerebralateryAftermodelingtemice wererandomlydividedintomodel group withoutinterventionsandinterventiongrouptreatedwithelectroacupunctureatthe acupointsBaihui (GV20),left Hegu (LI4),Neiguan (PC6), Zusanli (ST36),and Yanglingquan (GB34) 24h after surgery. The mice inte intervention group were housed individually incages withrunning wheels,andtheiractivitywasrecorded every 24h Neurological function scores of the mice were assessed on the 1st, 14th, and 21st days after modeling. Transdifferentiation of astrocytes inthetarget brain regions was observed using double immunofluorescence staining.Results Compared with thoseinthemodel group,the micereceiving eletroacupunctureandrehabilitationtrainingshowedsignificantimprovementof neurologicaldeficitsat14and21daysafter modeling.TheGFAPpromoteroftheAAV2/5vectorspecificalllabeledthelocal astrocytes,andcomparedwiththatthatinthemodel group,thenumberofAAV-positivecelscolabeledwiththeneuronal marker DCXsignificantlyincreasedafter14daysof electroacupunctureandrehabilitationintervention,andthenumberof AAV-positivecelscolabeledwiththeneuronalmarkerNeuNsignificantlyincreasedafter21daysofintervention.Concusion Inmicewithcerebralischemia,electroacupunctureandeabilitationtrainingcanpromotetransdierentiationofatrocytes intoneuronsintheischemicbrainregion,andtheeficiencyoftransdifferentiationispositivelycorelatedwiththe improvement of motor function.
