[Abstract]Vascular calcification is a common complication in patients with chronic kidney disease (CKD). With the progression of CKD,both the incidence and severityof vascular calcification graduall increase,which seriously affects the prognosis of patients.However,there are currently no clinically proven therapies specifically targeting vascular calcification for improvement. Klotho is aprotein possesing functions suchasantioxidant,antiinflammatory,and regulation of calcium-phosphate metabolism.It modulates multiple calcification-related pathways,including fibroblast growth factor-23,Runt-related transcription factor 2,Wnt/ β -catenin,phosphate transporters,and the receptor activator of nuclear factor KB,thereby inhibiting the formation of vascular calcification. Consequently,Klotho may bea promising novel therapeutic target for vascular calcification.This article reviews therole of Klotho in inhibiting vascularcalcification and its underlying mechanisms in CKD patients.
