Abstract:In order to explore the potential of cells as carriers for drug loading, a physical extrusion microfluidic drug loading device with strong comprehensiveness is proposed for the efficient loading of various drugs into cells. The device is composed of the fluid distributor at the front end and multiple parallel extrusion channels at the back end, which can ensure good drug loading effect while having high sample flux and low blockage. The working mechanism of the device and the effect of flow rate on the cell extrusion effect were elucidated by means of the numerical study, and the size of the surface strain was used as a characterization of the cell extrusion effect. The results show that during the extrusion process, the loading effect increases with the increase of the extrusion speed. The extrusion chip can disperse cells more uniformly into different extrusion channels, and reduce partial channel blockage caused by large differences in cell distribution in multiple channels and the difference in flow velocity distribution between multiple channels, thereby reducing the difference in extrusion effect.
