COX-2在口腔鳞状细胞癌增殖与侵袭中的作用研究

Abstract:ObjectiveTo explore theroleof cyclooxygenase-2(COX-2) in the proliferation,migration,invasionand tumorigenesis in vivo of oral squamous cellcarcinoma (OSCC)cels.MethodsTisse samples,including five casesof normal oral mucosaand fivecasesof OSCC,were colected postoperatively from the Department of Stomatology,Hainan Cancer Hospital.Immunohistochemistrywasperformed to detect the expression ofCOX-2in both groups.Celecoxiband COX-2 shRNA wereused toinhibit theexpressionofCOX-2inSCC9cells,andtheinhibitionefciencywasverified by Western blot assay.The proliferationabilityofcels was evaluatedbyCCK-8andcolonyformation assays.Themigrationand invasionabilitiesofcellsweredetectedbyscratchandinvasionassays.AsubcutaneousxenografttumormodelofSCC9cells in nudemice was established to observethe efect ofCOX-2 silencingon tumor growth.ResultsImmunohistochemistry showedthat COX-2 was mostly negative or weakly positive in normal oral mucosa tisse,but strongly positive in OSCC tissue.Compared with thecontrol group,both Celecoxib treatmentand COX-2shRNA transfectioncoulddown-regulate the expression of COX-2 protein in SCC9 cells ( P<0.05 ),and reduce the 0D450 value of cells, the number of clones,the scratch healing rateand the number of transmembrane cels.The xenograft tumor experiment in nude mice showed that with the extensionof tumorformationtime,the tumor volume inthe COX-2 silencing group decreased,andthefinal measured body weight was lower than that in the empty vector group ( P<0.05 ). Conclusion COX-2 is highly expressed in OSCC and participates intheproliferation,migration,ivasionandinvivogrowthof tumors.TheihibitionofCOX-2can weakenthe malignant biological behavior of OSCC cells.
