The effect miR-493-3p on high glucose-induced myocardial cell injury and apoptosis via the IncRNA CCAT2/TCF7L2 axis

Abstract:ObjectiveTo investigate the impact miR-493-3pon high glucose-induced cardiomyocyte injuryand apoptosis byregulating long noncoding RNA (lncCCAT2)/transcription factor7-like2(TCF7L2)axis.MethodsDiabetic cardiomyopathy(DCM)cellmodel(AC16)was established invitro with high glucose (HG,55mmol/Lglucose).Experimental grouping:thecontrol group,the HG group,the si-RNA negative control group,the si-CCAT2 group,theRNA mimics negative control group,the miR-493-3p-mimics group,the miR-493-3p-mimics + Lentivirus negative control group and the miR-493-3p-mimics +LV-CCAT2 group.Cell viabilityandapoptosisrate were detected byCCK8assayand flow cytometry.The levels lactatedehydrogenase(LDH) andaspartate aminotransferase (AST)incellculturemediumand superoxidedismutase (SOD) incels were detected bythe kit methods.Dual luciferase reporter gene assaywas used to verify the targeting relationship betweenmiR-493-3p and lncRNA CCAT2.The expression miR-493-3p,lncRNA CCAT2, β -catenin,TCF7L2,Caspase-3andCyclinD1genes were detectedbyRT-PCR.Westernblotassaywasusedtodetectthe expressions β -catenin (nuclear), β- catenin(Cytoplasm), TCF7L2, Caspase-3 and CyclinD1 proteins.ResultsCompared with the control group,miR-493-3p, OD 450 and SOD were decreased in the HG group,and levels lncRNA CCAT2, LDH, AST,apoptosisrate,the expresionlevels TCF7L2mRNAandproteinincreased.Up-regulating miR-493-3porknocking down the expression lncRNA CCAT2 can antagonize the above changes ( P<0.05 ). The double luciferase reporter gene experiment verified the relationship betweenmiR-493-3pand CCAT2and TCF7L2.Compared with the HG+miR-493-3pmimics group, the TCF7L2, β -catenin (nuclear),downstream Caspase-3 and CyclinD1 mRNA and protein expression were increased in the HG + miR-493-3p-mimics+LV-CCAT2 group( P<0.05 ).ConclusionmiR-493-3p can alleviate myocardialcellijuryandapoptosisunderhigh glucoseconditionbyinhibiting lncRNACCAT2expresion,andtereisa competitive relationship between the two regulation TCF7L2.
