Advances on ferroptosis in hematological malignancies

Abstract: Hematological malignancies are a group of heterogeneous diseases originating from the hematopoietic and lymphatic systems,characterized fundamentally by malignant clonal proliferation and differentiation arrest.Clinically,there are often significant challenges of poor prognosis and high risk of recurrence.Ferroptosis,an iron-dependent form of regulated cell death driven by lipid peroxidation, has been confirmed to be closely related to the pathogenesis, progresson,and therapeutic response of hematological malignancies. This article systematically reviews the molecular mechanisms of ferroptosis. It focuses on the research developments of hematological malignancies represented by leukemia,multiple myeloma,myelodysplastic syndromes,and lymphoma through the regulation of ferroptosis-related genes and pathways in order to provide a theoretical foundation for a deeper understanding of the disease process and the development of targeted treatment strategies.
