AbstractObjective:ToexplorethepotentialmechanismofYiqiHuoxueTongmaiFormulaforthetreatmentofreverse-dipper hypertensionbasedonnetworkpharmacologyMethods:ThebioactivecomponentsofYiqi HuoxueTongmaiFormulawerescreenedand theirtargetswerepredictedbyusingtheChineseMedicineSystemsPharmacologyDatabaseandAnalysisPlatform(TCMSP),AA TCM Database,PubChemdatabaseand Swiss Target Predictiondatabase.The DisGeNET,GeneCardsandOMIMdatabases were apliedtocollecterelevanttargetsofreverse-dipprhypertensionTheVenny2.1softarewasusedtomapthetargetsoftheYiqi HuoxueTongmaiFoulaandeverse-dipperhypertesionbtaintheommontargetsofrugsanddiseases,dratheVeiagra, constructthe"drug-component-ntersectiontarget"networkdiagramandtobtaintecoreactiveingredients.Theprotein-protein interaction(PPl)networkwasconstructedusingtheSTRINGdatabaseandCytoscape3.9.1softwaretoscrenandobtainthecore targets.TheDAVIDdatabasewasusedforgeneontology(GO)functionalannotationandKyotoEncyclopediaofGenesandGenomes (KEGG)pathwayenrichmentanalysisofcommontargetgenesfordrug-diseaseResuts:Atotalof109compoundsand1256targets werescreenedoutfromtheYiqiHuoxueTongmaiFormulaincluding1231targetsforreverse-iperhypertension13intesection targetsand11keytargets.TheresultsofGOenrichmentanalysisshowedtherewere1202biologicalprocesses,125cellar componentsnd225molecularfunctios.KGGenrichmentanalysisincuded189sinalingpathways.Thepotentialagetsiledin theinterventiooftiHouegaolafoveertesiodulin)tekin6r protein53(TP53),transcriptionactivator3(ST3)andtyrosinekinase(SRC).Teelatedsignalingpathwaysincludedteadaced glycationendproductsandteireceptors(AG-AGE)sinalingpathayinabeticomplicatios,tepoia-iduciblefacto-) signalingpatwaythcaciusigningpathayteposphatidyliositol3-kinase-proteininaseB(P3K-AKT)signalingpatayte mitogen-ativatedproteinkinase(MAPK)signaingpatwayap1signalingpathwaysprolactinsinalingpathwaysandrelainiing pathways.Conclusion:YiqiHuoxueTongmaiFormulafortreatingreverse-dipperhypertensionshowedthecharacteisticsandadvantages fmutipleomponents,utipeagetsndmutplepathwaysanditouldseeasanimportantresearchbasisforfurtrexploingthe active substances and mechanism ofaction of Yiqi Huoxue Tongmai Formula in treating reverse-dipper hypertension.
