AbstractObjective:Toinvestigatetheefectandrographolide(AD)onhighglucose(HG)inducedpyroptosismyocardialcelsby regulatingtheadenosinemonopospateactivatedproteinkinaseAKodikeeceptorprotein3(P3)ignalingpathayetos: Logarithmic phase H9c2 cells were divided into control group,HG group(50 mmol/L HG), HG+AD-L group 30mg/L AD+50mmol/L HG),HG+AD-M group 40mg/L AD+50 mmol/L HG),HG+AD-H group (50mg/L AD+50 mmol/L HG),and HG+ADH+AMPK inhibitor Compound C(CC )970μρ(50mg/↕ΔD+50mmol/↕HG+10μmol/L CC).CCK-8 method and Hoechst 33342/propidium iodide(Pl) stainingwereusedtodeterinetheproiferationactityandcellpyoptosisH9c2cell.Enzelinkedimmunosorbentassy) methodwasapliedtodeterminethelevels lactatedehydrogenase(LDH)andinflammatoryfactors.Reversetranscriptionpolymerase chainreactionqR-PCR)wasapiedtomeasurethemRNAexpressioncellpyroptosisfactor.WestemBlotwasapliedtodetermine the expression AMPK/NLRP3 pathway proteins and pyroptosis proteins.Results: 10-60mg/L AD was able to enhance the HG-induced proliferation activity H9c2 cells,andafter interventionwith 50mg/L AD,the proliferation activity H9c2 cels was the highest. Therefore,30,40,and 50mg/L AD were selected for subsequent experiments.Compared with the control group,the OD570 value and p-AMPK/AMPKintheHGgroupdecreased,whilethecellpyroptosisrate,DH,levelsinflammatoryfactors,mRNAandprotein expression pyroptosis factors increased( P<0.05 .Comparedwiththe HG group,the OD570 value and p-AMPK/AMPK in the HG+AD-L group,HG+AD-Mgroup,andHGAD-Hgroupalincreased,whiletheremaining indicatorsmentionedabovedecreased P<0.05) Compared with the HG+AD+H group,the OD570 value and p -AMPK/AMPK in the HG+AD-H+CC group decreased,while the remaining indicatorsaboveallincreased P<0.05 ).Conclusion:AD canalleviate HG-induced inflammatory damage to H9c2 cels,inhibit cell pyroptosis,and this effect may be related to regulating the AMPK/NLRP3 signaling pathway.
