Analysisofcomponentsmigrating to bloodandmetabolitesof Polygonumcuspidatum inrats with acute goutyarthritis

ABSTRACTOBJECTIVEToanalyze thecomponents migrating tobloodand metabolitesofPolygomumcuspidatum inrats with acute gouty arthritis (AGA).METHODs SD rats were randomly divided into blank group,model group and P. cuspidatum group ( |10g/kg ,byraw material),with 6rats in each group.Except for blank group,AGA model was induced in the remaining groups byinjecting potassiumoxonateand sodiumurate;meanwhile,theywereadministeredcoresponding drugsolutionsorwater intragastrically,onceaday,for1Oconsecutive days.Thehistopathological morphologyof thekne jointtisses inratswas observed;rat serum samples were collected,and the components migrating to blood and metabolites of P. cuspidatum were analyzed byusing UPLC-Q-Exactive-Orbitrap-MS. RESULTS Following the intervention with P. cuspidatum,the histopathological morphologyoftheknee jointsynovialtisueinAGAratsshowedsignificantimprovement,withreduced inflammatorycel infiltrationandhyerplasiandthepreservationofteoneyombikestructureintegrityInbothpositiveandnegativeiodes, a total of 67 chemical components were detected in the serum of rats from P. cuspidatum group,including 25 prototype components and42metabolites.The involvedcompoundtypes encompassed stilbenes,anthraquinones,naphthols,andflavonoids,among others.Temetabolicreactionsidentifiedincludedmethylation,acetylation,sulfation,andglucuronidationNotably,compounds suchas polydatin,resveratrolandemodinwerecapableofentering thebloodstreamintheirprototypeformsandundergoingin vivo metabolism.CONCLUSIONsCompoundssuch aspolydatin,resveratrol and emodinarelikelytobe theactivecomponents responsible for the anti-AGA effects of P.cuspidatum.
