AbstractDaning sun-cured tobacco is a species of good-quality yelow sun-cured tobacco in Hezhou, Guangxi,and it is the excellent raw material for cigarette production.However,it has extremely low resistance to black shank disease.This study investigated the response mechanism of Daning sun-cured tobacco to black shank disease by comparing the diferentially expressed genes and metabolites between healthy and black shank-infected leaves through transcriptomic and metabolomic analyses.The results showed that a total of 7 194 significantly diferentially expressed genes were identified through transcriptomic analysis,and 198 significantly diffrent metabolites were detected through metabolomic analysis.The combined transcriptomic and metabolomic analysis revealed that : ① Plant hormone signal transduction and phenylpropanoid biosynthesis pathways played important roles in the defense of Daning sun-cured tobacco against black shank infection. ② 二 was speculated that metabolic pathways such as aminoacyl-tRNA biosynthesis,cyanogenic amino acid metabolism,valine,leucine and isoleucine degradation,cysteine and methionine metabolism,and arginine and proline metabolism were related to the response of Daning sun-cured tobacco to black shank stress.This study elucidated the major gene transcriptional regulatory networks and metabolic pathways of Daning sun-cured tobacco in response to black shank infection,which could provide a theoretical basis for research on improving the resistance of Daning sun-cured tobacco to black shank disease.
