Study on Extraction of Polysaccharides from Cistanche deserticola by Different Processes and Their Antioxidant Activity

Abstract: Water extraction method,enzymatic extraction method and ultrasonic-assisted extraction method are used to extract polysaccharides from Cistanche deserticola. The extraction yield is compared,and the extraction process is optimized. Meanwhile,the antioxidant activity of Cistanche deserticola polysaccharides is determined. The results indicate that among the three methods,ultrasonic-asssted extraction method has the highest extraction yield. Optimization of extraction process is performed on this method. Using single factor test combined with response surface method,the solid-liquid ratio,ultrasonic extraction time and ultrasonic extraction temperature are selected for comparative analysis.Finally,it is determined that when solid-liquid ratio is 1:100 (add 5.0g Cistanche deserticola powder into 500mL distilled water),ultrasonic extraction time is 40min ,and ultrasonic extraction temperature is 48°C ,the extraction yield is the highest of 7.49% . At this time, the DPPH radical scavenging rate of polysaccharides extracted from Cistanche deserticola is 84.27% of that of vc ,and the ABTS radical scavenging rate is 73.95% of that of vc
