Abstract:【Objective】Toanalyze thegeneticdiversityandgenetic structureofdiferentgenerationsofChinese firbreeding populations,econstructthegeneticpedigreeofbreedingparentsfromeachgenerationtoguidetheselectionandmatingofbreeding parents,andestablishacoregermplasmofChinesefir.【Method】Atotalof388Chinesefrbredingclonalparentsfromdierent generations(74fromtheinitialgeneration,70fromthefirsgeneration,107fromthesecondgeneration,andl37fromthethird generation)wereusedasmaterials.GeneticdatawereanalyzedbasedonESTSSR,andtheoverallevelofbreedingpopulations was reflectedbythepolymorpismandgeneticdiferentiationparametersof16ociGeneticdiversityparameters,geneticvariationand geneticdiffeentiationecentsreusdtoaaletgeneticdivesitylevelsofCineefrreingpoplatiosacodient generations.Clustering analysisand population structure analysis wereconductedtodetermine population groupings and genetic pedigreesofbreeding populations.Coregermplasmwasseletedbasedontheprincipleofmaximizingalleles.【Result】1)Atotal of 640 polymorphiclociwereamplifiedwith16primerpairs,withanaverageof40allelesperlocus.The meanefectivenumberof aleleswas16.25,theShanonindexwas2.942,themeanpolymorphisminformationcontent(C)was0.942,andthemeanfation indexwas0.97indicatinghighpolymorpismamongpopulations.TeoverallgeneticdiversityofteCinesefrredngoplations washighethantatofteinitialgeneratio,withecondandthidgeneratiosehbitinghigherdiversitytantheovealevel; 2)Thefourbreedingpopulatnseredividedintoteediferetgroupsithindividualsgroupedaccordgtomateralcaracteistic andgeneticrelationships;3)Atotalof127coregermplasmswereselected,andtheconstructedcoregermplasmbankretained 100% of thealeles.Thefectivenessoftecoregermplasmbankwasverifedthroughageneticparametertest.【Conclusion】Tedieent generationsofesefireedingpoplatioseiedggeticdversitydlowgneticderean,dicatigtatdace generationbreedingefectivelymaintains geneticvariation withinpopulations.Thereconstructedpedigreemapofbreedingparents providesasientificbasisforteatoalseectionndrossbreedingofeliteparentsMoreoverthidentifedorcolectietains 100% oftheallelicdiversityoftheorigialgermplasmandshowsstrongrepresentativeness,ayingasolidgeneticfoundationforthe futureelectioofsuperiorarieties,fentutilationofgerplasesoues,andsustaiabledevelopmentofreedingolations.
