AbstractObjective To investigate the indolocarbazole alkaloids produced by marine-derived Streptomyces sp. OUCMDZ-2582 and their antibacterial activity. MethodsA1 liquid culture medium was used to ferment the Streptomyces strain.The fermentation broth was extracted by ethyl acetate (EtOAc). Guided by the characteristic ultravioletabsorptionof indolecarbazole,the indolecarbazole alkaloids inthe fermentation products were isolated by column chromatographyover positive and reverse phase silica gel,Sephadex LH-2O,aswellassemi-preparative high-performance liquid chromatography (HPLC) to give pure compounds. The structures of the compounds were elucidated through theanalysisofLC-MS,NMR,andoptical rotation,etc.The antibacterial activity was assyed by microbroth dilutionmethod.ResultsNine indolocarbazole alkaloids were purified and identified as staurosporine (1),3'-N-acetsturooine (2),3--burospie(3),A (4),trtocboleC(5),b, 3'-N-acetylholyrineA(7),3'-N-acetyl-3-ydroxyholyrineA(8),andK25c(9),respectively.Teantibacterialaciviie for compounds 2and 3 against Vibrio parahaemolyticus and compound 3 against Vibrio alginolyticus were observed, each at a minimum inhibitory concentration(MIC) of 32μg/mL . Conclusion Streptomyces sp.OUCMDZ-2582, isolated from the Yellow River Delta,is a prolific producer of indolocarbazole alkaloids. Compounds 2 and 3 displayedantibacterial activities.
