高效液相色谱法测定砂生槐种子中3种生物碱含量的方法研究

【Abstract】ObjectiveTo optimize the method for determining alkaloids in Sophora ammosa,and to determine thecontentof mainalkaloidsin Sophoraammosa byhigh-performance liquidchromatography (HPLC). MethodsUsing C18 column,methanol and sodium phosphate bufer salt (PH8)as mobile phase,detection wavelength 210nm ,column temperature 35°C ,flow rate as chromatographic conditions,respectively, the content of 3 alkaloidsandsamples were determined,and thestandard curveand RSDof the method were evaluated.ResultsThe linear relationshipof the method wasgood inthestandard curve rangeof alkaloid concentration from O.54 to 60.00μg/ml ,and the absolute values of correlation coefficients were all greater than 0.992.The concentrations of low,medium and high alkaloids (6.O,12.O and 30.0μg/ml )weredetermined.The average recovery ranged from 84.4% to 97.4% . The content of oxymatrine was 2.644mg/g ,sophorine 0.095mg/g and matrine was 0.047mg/g .Conclusion The optimised HPLC method can detect the content of 3 alkaloids in thesamples,which hastheadvantagesofhigh eficiencyandsaving time,andissuitable for the detection of alkaloidsin themulberryfruit.
