Simultaneous Determination of Seven Components in Curcumae Radix by High Performance Liquid Chromatography

Abstract:ObjectiveTo establish a method forthe simultaneous determinationof sevencomponents(aerugidol, demethoxycurcumin,curcumin,curcumenol, isocurcumenol,germacrone,and furanodiene) in Curcumae Radix based on high performance liquid chromatography (HPLC).Methods HPLC analysis was performed using a Waters Symmetry C18 column ( 4.6mm×250mm , 5μm ).Agradient elution system consisting of acetonitrile(A)and- 0.2% (204号 phosphoric acid aqueous solution(B) was used as the mobile phase.The flow rate was O.8, ,the detection wavelength was set at 204nm ,the column temperature was maintained at ,and the injection volume was 10μL .Results The seven components in Curcumae Radix were well separated. The linear ranges for aerugidol, demethoxycurcumin,curcumin,curcumenol, isocurcumenol,germacrone,and furanodiene were 4.84-48.36,0.08- 3.36,0.16-3.25,3.20-32.00,0.32-3.22, 1.61-32.24,and 0.32-6.48μg⋅mL-1 ,respectively.All components exhibited excellent linear relationships ( r⩾0.9995 ). The average recovery rates ranged from 96.37% to 108.47% ,with 1 RSD⩽2.14% . In the analysis of 22 batches of Curcumae Radix samples,the average contents of the aforementioned seven components were 308.39,14.88,25.30,328.72,21.83,152.00, and46.20μg∙g-1 ,respectively. Conclusion This method isaccurateandreliable,suitable for thequalityevaluationof Curcumae Radixand the quantitative determinationof itssevencharacteristiccomponents.Itcan provideascientific basis forthequalitycontrolof Curcumae Radix.
