花生品种萌芽期耐盐性综合评价及QTL定位

Abstract:In this study,12 peanut varieties were subjected to salt stress treatments with NaCl solutions at seven concentrations(0,50,75,85,100,125,and 150mmol/L ),and 85mmol/L was identified as the suitable concentration for testing salt tolerance at the germination stage.Using 85mmol/L concentration,the salt tolerance identification was further performed on 88 peanut varieties from Huang-Huai-Hai region,deploying relative germination energy,relative germination rate,relative shoot length,relative shoot weight as salt tolerance evaluation indicators. Comprehensive evaluations were conducted using principal component analysis, membership function,and systematic clustering.Based on salt tolerance,the 88 peanut germplasms could be divided into five major groups: highly salt-tolerant type,salt-tolerant type,intermediate type,salt-sensitive type,and highly salt-sensitive type.Two highly salt-tolerant varieties including Fenhua 13 and Huayu 910,and two highly salt-sensitive varieties including Huayu 9144 and Longhua 7,were obtained.Furthermore,genetic analysis of salt tolerance was caried out on a genetic population of 2Oo families which were derived from Huayu
44(salt-tolerant) ×DF12 (salt-sensitive) and genotyped by whole-genome resequencing. Based on the highdensity genetic map comprising 20 chromosomes,QTL mapping identified four QTLs under salt stress conditions,located on chromosomes A10 and B10,with phenotypic variation explanation rates ranging from 4.49% to 4.81% . This study provides germplasm materials and a theoretical basis for the identification and breeding of salt-tolerant peanut varieties and the mining of salt-tolerant genes.
