Investigation on causal relationship between 473 gut microbiota and pertussis based on Mendelian randomization method

【Abstract】ObjectiveToinvestigate causal relationship between 473gut microbiotaandpertusis by genome-wide Mendelianrandomization method.MethodsGut microbiotadata were derived from genome-wideassociation study(GWAS), whichincludedatotalof5959samplescovering 473gutmicrobiotaspecies.Pertussisdata wereextractedfromtheFinnGenR9 database(147casesand332343controls).Inverse variance weighted(IVW)methodwasusedforbidirectionaltwo sample Mendelian randomization analysis to evaluate the causal effect of gut microbiota on pertusis,and Cochran's Q test,MR-Egger intercepttest,andMR-PRESSO wereusedtoexamineheterogeneityandpleiotropy.ResultsAtotalof 2Ogutmicrobiota were identifiedascausalllinked topertusis.Among them,11microbiota exhibitedprotectiveefects,withUBA7177showingthe strongest protection (OR=0.1404,95% CI : 0.0240 - 0.8214, P=0.0294 );9 species increased the risk of pertussis,with Lawsonella spOO2161175 presenting the highest risk( OR=15.2697 ,95%CI: 2.3443-99.4593,P=0.0044). Conclusion
Acausal relationship exists between gutmicrobiotaandpertusis,and targeted modulationof specificmicrobiotamayffer novel strategies for the prevention and treatment of pertussis.
