Study on the Antibacterial Effect and Mechanism of Dipsacus Asper Extract Based on Ionic Liquid and Microwave-assisted Method

Abstract:Using Dipsacus as the raw material,the extract was obtained through ionic liquid and microwave-assisted extraction technology.Antibacterial experiments and active component analysis of the extract were conducted,and the antibacterial mechanism was discussed.The results show that the Dipsacus extract exhibits antibacterial effects against Escherichia coli( E . coli),Staphylococcus aureus (S. aureus),and Salmonella paratyphi β ( S .paratyphi β ),with its inhibitory activity being concentration-dependent. The inhibition zone diameters for E . coli,S.aureus,and S . paratyphi β are 27 mm,39 mm,and 19mm ,respectively. The minimum inhibitory concentration (MIC)and minimum bactericidal concentration(MBC) against S. aureus are determined to be 3.1250mg/mL and 6.250 0mg/mL , respectively. For E : coli,the MIC and MBC values are 3.1250mg/mL and 6. 250 0mg/mL , respectively. For S :paratyphi β ,MICis 6.250 0mg/mL and MBC is 12.500 0mg/mL . Chromatographic analysis reveals the presence of antibacterial components in Dipsacus extract,including chlorogenic acid,caffeic acid,and asperosaponin VI. Morphological observations suggest that the antibacterial mechanism may involve disruption of bacterial cell wals and membranes.These findings provide valuable references for the development of antimicrobial agents from Dipsacus.
