Abstract:The gymnosperm cedar(Cedrus deodara)isawind-pollnated plant,and exhibitsa significantlyslower proces pollen germination and polen tube growth than that angiosperm(e.g.,maize).Starch is the mainenergy source for polln germination inwind-pollnatedplants.Toreveal thecharacteristics starch granulesincedar pollenand their changes during pollngermination,thepollens cedarand maize wereusedas materials tocompare thechangesstarch granulesandtheireffectsduring polengerminationbyphysiologicalandbiochemical,cytologicalandproteomic aproaches.The results were as follows:(1)Compared with mature maize pollen,the starch content in cedar polen was lower,buttheamylose proportionwas higher.(2)Starch granules in cedar pollen were iregularand cobblestone-like, whereas those in maize were short and rod-shaped.(3)The activities α -amylase and β -amylase,and the types and activitiesisoenzymes weremuch lower than thoseinmaizepolen.(4)Starch granulesinmaturecedarpolen were primarilydistributed in thecytoplasm around theinner pollenwall,and gradually moved intopollentubeduring pollen germination.Meanwhile,thetypesandactivitiesstarch isoenzymes increased,andthedegradationstarch increased, thereby producing more soluble sugar to provide energy forpollen germination.Thisresearch provides new clues for understanding the reasons for the slow pollen germination and pollen tube growth gymnosperms.
