Association between molecular phenotypes and clinicopathological characteristics in gliomas: an immunohistochemical analysis

Abstract: Objective To investigate the expression patterns of immunohistochemical markers in glioma tissues and their correlation with clinicopathological parameters,providing a basis for molecular clasification and prognosis assessment.Methods Twenty-one glioma cases were collected. Immunohistochemistry was performed to detect the expression of molecular markers,including glial fibrillary acidic protein (GFAP), isocitrate dehydrogenase-1 R132H mutant protein (IDH-1 R132H),Ki-67 antigen (Ki-67),S-100 protein (S-100),synaptophysin(Syn),and tumor protein p53(p53).Statistical analysiswas conducted in conjunction with clinical data and WHO grading. Results The study cohort was predominantly composed of high -grade gliomas (WHO grades I~N , 66.7% ).Glioblastoma(9 cases, 42.9% )was the most common subtype.The incidence was higher in patients aged 46 to 61 years (38.1%) )and ⩾61 years (33.3%) : Molecular marker analysis revealed high positive rates for GFAP ( 90.5% )and S-100( 85.7% ). The Ki-67 proliferation index ranged from 2% to 40% . An aberrant p53 expression rate of 55.6% was observed. A Syn positivity rate of 66.7% suggested a tendency toward neuronal differentiation in some of the tumors. Among the 14 cases tested for IDH-1,13( 92.9%) were wild-type,and only1 (7.1%) was mutant. Conclusion The gliomas in thiscohort were primarily IDH wild-type and clustered in themiddle-agedand elderly patients exhibiting features of high proliferative activity(elevated Ki-67 index)and aberrant p53 expression. This molecular profile helps define a more aggressive glioma subtype and holds referential value for its clinical diagnosis and prognosis evaluation.
