【Abstract】ObjectiveThrough the study of the expression and clinicopathological significance of the TrNA-derived fragmenttRF-Glu-TTC-2 in PCa,theposibilityof it as a new target in the diagnosis and treatment of prostate cancer(PCa) was explored.MethodsThree pairs of fresh PCa tisues and normal prostate tissues frominpatients intheAfliated Hospitalof Yangzhou University from January2018 to December2O21 were selected.Shanghai Kangcheng BiotechnologyCo.,Ltd.was entrusted to detect thetRFand tiRNAexpression profiles of PCa through tRF and tiRNA microarrays.The tRF-Glu-TTC-2 with the maximumupregulation multiple wasselectedas theresearch objectand verified byreal-time fluorescence quantitative polymerase chain reaction (qRT-PCR).The probe was designed based on the detected tRF-Glu-TTC-2 nucleotide sequence.The expression of tRF-Glu-TTC-2 in PCa tissues and the correlation between its expression and clinicopathological parameters were detected by RNA in situ hybridization (RISH) technology.ResulttRF&tiRNA expresson profilesand qRT-PCR verificationexperimentsshowed that the expresion level of tRF-Glu-TTC-2 inPCaincreased significantly.Its nucleotide sequence is 5′ -TCCCACATGGTCTAGCGGTTAGGATTCCTGG- 3′ ,choose the nucleotide sequence is thecore of the oligonucleotide probe design,lineRISH tests revealedagency-Glu -TTC-2 inPCacells positive expression in thecytoplasm.The high expresson oftRF-Glu-TTC-2 was closelyrelated to the tumor sizeof PCa( χ2 =8.122,P=O.0O4),but not related to age,presence or absence of lymph node metastasis,or TNM clinical stage (all P>0.05 ). Conclusion tRF-Glu-TTC-2 may be a new oncogene.Its expression in PCa tissues is closely related to tumor size and Gleason score,which provides anew idea for the clinical diagnosis and treatment of PCa.
