AbstractObjective:Toinvestigatethemechanismbyhichgastrodininhibitspyroptosisinmiddlecerebralarterylusion(MCO)rats throughthelongnon-codingRNA(IncRNA)matemalyexpressdgene3(Meg3)/miR-485/absentinmelanoma2(AIM2)pathwayvia exosomes,therebyprovidinganexperimentalbasisforearlyinterventionwithtradionalChinesemedicineduringtheacutestageof cerebral infarction.Methods:SDatsweredividedintocontrolgroup,shamoperationgroup,modelgroup,andgastrodingroup.emodel group and the gastrodin group were subjected with MCAO surgery.Gastrodin at a dose of 100mg/(kg⋅d) wasadministered intraperitoneally.SD ratsweresacrficdondays13and7.Teischemichemisphereandserumwerecolected.Exosomeswereextractedfromtheseru. Thelevelsof NOD-likereceptorprotein3(NLRP3)andAIM2intheischemichemispheresweredetectedbyWestemblotingand immunohistochemistryThelevelsofNLRP3andAIM2 mRNA,as wellasIncRNA Meg3andmiR-485,intheischemichemispheresof ratsineachgroupweredetectedbyquantiativeeal-timepolymerasechainreaction(qPCR).Exosomeswereisolatedfromperipheral plasmaandthelevelsofncRNAMeg3andmiR-485weremeasuredbyPCRResults:WestemBlotresultsshowedthatthecontentsof NLRP3and AlM2 intheischemic hemisphere of gastrodin group were lower than those inthe model groupe (P<0.05) .The immunohistochemicalresultsshowedthesametrend.TheqPCRresultsshowedthatthemRNAlevelsofAIM2intheischemic hemisphereofratsintegastrodingroupoestrdand7tdaystercerebralinfarctionwereallowerthantoseinodel group P<0.05 or P<0.01 ).The IncRNA Meg3 level wasalso lower than that in the model group( P<0.05 or P<0.01 ),while themiR-485 level was higher than that in the model group P<0.05 or P<0.01 ).The content of IncRNA Meg3 in serum exosomes of the ginsenoside group was lower than that of themodel grouponthe1st,3rd,and7thdaysaftercerebral infarction P<0.05 or P<0.01 ),whilethecontent of miR-485 was higher than that of the model group( P<0.01 ).Conclusion:Gastrodin probably inhibits pyroptosis in MCAO rats by regulating the IncRNA Meg3/miR-485/AIM2 pathwayviaexosomes.
