棘胸蛙ISG56和ISG58基因结构特征及其对细菌性感染的免疫应答

Abstract:[Objective]This studyaimed to elucidate theresponsecharacteristicsofinterferon-stimulated genes ISG56 andISG58intheQuasipaa spinosaunderbacterial infections,therebyprovidingatheoreticalbasis fordiseaseprevention,control,and ecofriendly breeding offrogs,andenriching the immunobiological research data of amphibians. 【Method]The sequences of ISG56 and ISG58 genes were cloned,and the bioinformatic analyses were conducted using online software such as BLAST,SMART,TPRpred,and MEGA 4.0 to investigate the change of ISG56 and ISG58 expressions under lipopolysaccharide stressand Aeromonas hydrophila infection.【Result]The total lengthsof theopen reading frames (ORF)of ISG56and ISG58 genes were1416 bpand1461bp,encoding 471and486 amino acid residues, andproteins encodedbythegenes included9and8TPRdomains whose lengths were34aminoacidresidues;their tertiary structures were helix-loop-helix.Phylogenetic treeof ISG56and ISG58 amino acid sequences showed that,noclear boundary existed between the amino acid sequences of ISG56and ISG58 except for freshwater fishes and amphibians.The ISG56and ISG58 genes showed stable expressions intissues suchasspleen,liver,and kidney,but with some difference among tisues.After intraperitoneal injectionofLPS to healthy Quasipaa spinosa,theresponse intensitiesof the ISG56 and ISG58 genes in were not high in liver,spleen and kidneytisues,while the primary response was observed in liver tisues.The healthyQuasipaa spinosa,after infectedwithAeromonas hydrophila,exhibited mildresponsesof theISG56 and ISG58gene expressions inthreetissues ofliver,spleen and kidney;specificallyin livertherelative expressionof the ISG58 gene at 24h post-injection was significantly up-regulated by 69.3-fold compared to that at 0h post-injection (P<0.05) ;such effect was concentrated between 12 and 48 h post-injection.【Conclusion】In Quasipaa spinosa,the ISG56 and ISG58 genes show high expressions mainly in immune tissues.The proteins encoded by the genes contain 9 and8complete TPR domains,respectively;these proteins are important for theimmune response to bacterial infections and exert anti-inflammatory effects via non-interferon-dependent pathways.
