Abstract:TheJAZ(jasmonate ZIM-domain)isarepressr protein in the jasmonicacid signaling pathway,inhibiting the expresionof early jasmonicacid response genes by interacting with related transcription factors.To elucidate he characteristicsandactivityofthe HbJAZ1.Ogene promoter,this study used the rubbertree variety‘CATAS7-33-97’as material and analyzed the cis-acting elements and expression activity of the HbJAZ1.0 gene promoter through PCR amplification,PlantCARE software,and Arabidopsis thaliana transgenetic techniques.Theresults wereas follows:(1) A total of 1 5Ol bp promoter sequence upstream of the HbJAZ1.O gene was isolated from the genomic DNA of the rubber tree.(2)Several keycis-acting elements hadbeen identified within the HbJAZ1.O gene promoter,including ABRE for abscisic acid,TCA-element forsalicylicacid,aswellas ACAT-motif,Box 4,G-box,GT1-motif,TCCC-motif,TCTmotif and chs-CMA1a for light responsiveness.(3)A plant expression vector containing the GUS reporter gene under the control of the HbJAZ1.0 gene promoter had been constructed and transformed it into A. thaliana using Agrobacterium tumefaciens-mediated transformation. Histochemical GUS staining demonstrated extensive expresson of the HbJAZ1.0 (20 genepromoter throughoutvarious tissuesofArabidopsis thaliana,including rots,stems,leaves,flowers,andfruits.In conclusion,theresults indicate the HbJAZ1.O gene promoter has a constitutive expresionactivityand might beregulated bylightand various plant hormones.Theresults ofthestudy maylaythe foundation forfurther studyonthe functionof HbJAZ1.0 gene.
