Sequence analysis of PP2C2 gene in Artemisia desertorum and its expression pattern under drought stress

Abstract:Protein phosphatase 2C(PP2C)play important roles in growth and development,cellcycle regulation, signaltransduction,andresponses toenvironmental stressThisstudyfocusedonArtemisiadesertorum,ahighlydroughttolerant and multifunctional plant native to arid desert regions,to investigate the functional role of PP2C2 gene in drought tolerance.Using A.desertorumas theexperimental material,the PP2C2 genewasamplified,itssequence was subjected to bioinformaticanalysis,and itsproteinstructurewaspredicted.Additionally,theexpression pattrnsof PP2C2 under various water stress conditions were analyzed,and the regulatory role of the PP2C2 gene in drought stress responses was elucidated. The results were as follows:(1)The PP2C2 gene in A. desertorum contained an open reading frame(ORF)of1 404 bp,encoding a protein of 467amino acids.Sequence alignment demonstratedthatthe gene shared high homology with PP2C genes from other plants,such as Erigeron canadensis.(2) Bioinformatic analysis revealed that the PP2C2 protein was localized in the nucleus,no signal peptide,and was classified as a non-secretory protein.Its secondary structure predominantly consisted of random coils,with strong hydrophilicity,no transmembrane helical region,and multiple potential modification sites.(3)Quantitative real-time PCR(qRT-PCR)analysis showedthat the expression of the PP2C2 gene was responsive to varying levels of drought stress. Under drought conditions,its expression exhibitedanoverallup-regulation trend,whilerehydrationledtoasignificantreductioninexpression levels.This study preliminarily reveals that the PP2C2 geneplays a significant role in the drought tolerance mechanism of Artemisia desertorum,offeringatheoreticalreferenceforfurtherunderstandingof itsdroughtresistancemechanismand potential application in genetic engineering.

沙蒿(Artemisiadesertorum)为菊科(Asteraceae)蒿属(Artemisia)的一种多年生野生灌木,在中国大部分地区均有分布,其中以我国北方和西北地区较为常见,该植物主要生长在荒漠和半荒漠等干旱地区,根系发达,表现出良好的耐旱能力。(剩余18487字)
